TAKFLAM PLUS TABLET is formulated by using combination of ACECLOFENAC POTASSIUM and PARACETAMOL one NSAID with an analgesic and antipyretic. Mechanism of action of both the ingredients is different but when used in a combination it significantly gives grater effect than individual. ACECLOFENAC POTASSIUM gives anti-inflammatory effect and PARACETAMOL gives analgesic effect.

ACECLOFENAC POTASSIUM is multi-functional drug as anti-inflammatory it inhibits the both leukocyte migration and COX-1 and COX-2 enzyme which further inhibit the synthesis of prostaglandins cause of inflammation. As prostaglandins synthesis inhibits the prostaglandins sensitize pain receptors directly inhibit which shows the analgesic effect. Antipyretic effect explained by its action on hypothalamus as a result peripheral dilation occurs, increased cutaneous blood flow and subsequent heat dissipation.

As analgesic PARACETAMOL thought to act on central nervous system, reduces pain by inhibiting COX enzymes such as COX-1, COX-2 and COX-3. Unlike NSAIDS it does not show any kind of action on peripheral tissues that's why is not act as anti-inflammatory. Antipyretic effect of PARACETAMOL is explained by the direct effect of heat-regulating centre of hypothalamus as a results peripheral dilation and sweating occurs which cause heat dissipation.
